The History of Stenabolic (SR9009)

Stenabolic has a long and storied history, dating back to the early 2000s when it was developed by Professor Thomas Burris of the Scripps Research Institute. It was initially created as a treatment for seniors with age-related muscle loss.  Stenabolic aimed to be a synthetic drug that was created in the laboratory in an effort to specifically target and activate the receptors located in muscle tissue.

What is Stenabolic

Stenabolic is a SARM that binds to Rev-ERBα, which influences the body’s metabolic activity. When bound, Stenabolic increases the number of mitochondria in the cells.

Stenabolic Circadian Rhythm

Synthetically produced REV-ERB ligands have been demonstrated to have an effect and in essence change the circadian behavior in mice and to change the circadian pattern of core clock gene expression in the hypothalamus area of the brain. This was also observed in a variety of metabolic genes present in the liver tissue, skeletal muscle, and adipose tissues, resulting in an increase in energy expenditure possibilities[1].

When a REV-ERB agonist treatment was administered to diet-induced obese mice, the obesity level decreased as fat mass was lowered, and dyslipidemia and hyperglycemia were improved. These outcomes show that synthetically produced REV-ERB ligands that target the circadian rhythm might be beneficial for treating both sleep problems and metabolic illnesses.[1]

SR9009 Effects on Endurance

SR9009 administered to mice resulted in them being able to run longer distances and for longer periods than normal mice [2]. Overexpression of Rev-erb-α in vitro has been shown to boost the amount of mitochondria and better its respiratory capacity. In addition, overexpression or pharmacological activation of Rev-erb-α in vivo tests resulted in an increased exercise capacity- resulting in better endurance. This suggests that Rev-erb-α may one day be used as a pharmacological target to increase muscle oxidative function by regulating the gene networks [2].

SR9009 Effects on Cardioprotection

A study on mice low on REV-ERB demonstrated significant cardiac myopathy after transverse aortic constriction compared to the wild-type control mice group, indicating the significance of REV-ERB in protecting the heart from pressure overload. Interestingly enough, Stenabolic was also able to provide cardioprotective effects in REV-ERBα/β cardiac-specific double knockout mice (cDKO) mice.

Also, when administered at peak or trough points of REV-ERB expression, Stenabolic had similar levels of cardioprotection, implying that possibly some cardioprotective effects exist outside of REV-ERB. These results further demonstrate that deletion of REV-ERB accelerates negative cardiac remodeling in response to pressure overload and illustrates the independent cardio-protective aspect of Stenabolic with regard to pressure overload [3].

SR9009 effects on Lungs

Another study conducted in vitro showed that pharmacological activation of Rev-Erbα with Stenabolic can reduce the inflammatory body response in lung injury by suppressing TLR4-induced NF-κB activation. SR9009 usage was useful in providing positive effects on the metabolism, as it could ameliorate acidosis and hypoxemia by lowering the arterial PaCO2 and improving arterial PaO2, SO2, HCO3 –, lactic acid concentration levels, and blood PH levels. These results show that Stenabolic can reduce sepsis-induced lung injury and suggest targeting Rev-Erbα may be a promising strategy for preventing and managing acute lung injury  [4].

A different study dealing with investigating the anti-tumor effects of SR9009 in the lungs demonstrated that SR9009 is especially bad for both chemo-sensitive and chemo-resistant small cell Lung cancer cells. REV-ERBα was shown to be involved in the antitumor effect of Stenabolic. Also, the core autophagy gene Atg5 was demonstrated as a direct downstream target of REV-ERBα, which was suppressed by the REV-ERB agonist. The interaction between REV-ERBα and this autophagy gene prevented autophagy activity, thereby leading to Stenabolic cytotoxicity in SCLC cells [5].

SR 9009 and Energy Metabolism

LAst but not least, one study demonstrated that the administration of the REV-ERBα agonist Stenabolic has been shown to have an effect on energy homeostasis in animals [6]. This study on goldfish showed that SR9009 inhibited feeding and decreased body weight and length gain. In addition, the abundance of ghrelin mRNA was lowered in the intestine while that of leptin-aI increased in the liver tissue. SR9009 also was shown to decrease plasma glucose levels coupled with higher glycolysis and lower gluconeogenesis rate in the liver, while lipid levels and lipogenic enzyme activities were reduced too. Interestingly, the energy expenditure by locomotor activity remained unaffected. These results point towards REV-ERBα activation from Stenabolic promotes a negative energy balance by reducing energetic inputs and regulating lipid and glucose metabolism [6].

Stenabolic Side Effects 

Some research subjects administered Stenabolic have anecdotally shown side effects such as headache, fatigue, or nausea. These side effects have tended to be mild and disappear over time on their own.

Cardarine vs Stenabolic

Cardarine and Stenabolic are different in a few ways. Cardarine has a much longer half-life than Stenabolic. It is disputed whether Cardarine has a safer profile than Stenabolic, more research is needed to confirm what the answer is.

Stenabolic And Cancer 

There is no clear answer as to whether Stenabolic may promote cancer or not. Some research suggests that it might be a bad compound and could lead to the development of cancer, while other research suggests that it does not. Overall, it is inconclusive and more trials need to occur before there is a definitive answer on whether the compound causes cancerous behavior.
