Epitalon, also is commonly known by its other name Epithalon, is a short, four-amino acid synthetic polypeptide chain derived from epithalamin that activates the natural production of the telomerase enzyme and stimulates melatonin secretion from the human pineal gland [1]. Epithalamin in vivo helps to regulate cell replication and the anterior pituitary gland's correct functionality.

Epitalon was originally discovered by the Russian-born doctor and bioengineer Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology and has been studied for well over 35 years. It is praised by many anti-aging-related gurus and regarded as a potential fountain of youth. Studying mice and rats has shown that Epitalon may improve the condition when suffering from age-related diseases, skin health, and appearance, increase resistance to emotional stress, stimulate the production of new cells and rejuvenate muscle cells, and increase lifespan by delaying age-related changes in the reproductive and immune systems [13].

The above-described effect may be increased in individuals who do not have naturally long telomeres. Epitalon treatment also displays a significant effect on certain types of cancerous cells, infections, and DNA regulation [1].

DNA Protection

The Epitalon peptide has been shown to exhibit antioxidant behavior in the body by way of assisting in the elimination of free radicals.  Experiments done on somatic cells show that Epitalon peptide treatment activates an enzyme called telomerase and stimulates the natural production of telomerase [1].

 As commonly known, Telomere shortening is related to aging and Telomerase is one of several naturally occurring components that are considered the potential fountain of youth because it may stimulate the production of new cells and rejuvenate old ones. Telomerase enzyme activation leads to an improvement in cell overall general health, an increase in telomeres length, a decrease in errors contained in a strand of DNA when new DNA is copied, and protection of chromosomes and DNA from damage for example from free radicals [2, 3].

Studies conducted show that Epitalon interacts directly with DNA to increase the expression of certain genes, specifically regulating the CD5, IL-2, MMP2, and Tram1 genes and the natural production of telomerase[4,13,12].

Anti-tumor Effects

In studies conducted on rats, Epitalon has been shown to improve muscle cell health and decrease tumor growth rate and speed [5]. In In several studies Epitalon treatment was also shown to reduce tumor growth, and also to prevent metastasis which is the spread of cancer to other healthy cells and organs [6, 7]. Due to these interesting findings, Epithalon is currently being further researched as a potential treatment for Her-2/neu positive breast cancers, leukemia, and testicular cancers [8, 9].

Sleep Quality

Research conducted on rats has shown that Epitalon impacts circadian rhythm through both the formation and discharge of melatonin by relating to two proteins, pCREB transcription protein and arylalkylamine-N-acetyltransferase (AANAT) [10].  These two genes contribute to the normal production of melatonin in the body and the release of melatonin from the pineal gland. A monkey study suggests that Epitalon peptide therapy regulates melatonin release to normality after its release has been affected due to multiple reasons [11]. Improvements in circadian regularity cycles and deeper, better sleep have been known to increase skin health and overall general appearance, stimulate the production of new muscle cells, regulate the anterior pituitary gland function, increase cell replication, and promote resistance to emotional stress and age-related diseases. Sleep is also thought by many health experts as the natural fountain of youth and critical to muscle cell health, the production of new cells, and life extension [13].

Side Effects

Epitalone therapy has yet to show any significant side effects in human and animal studies however this does not mean that the use of Epitalon peptide is safe, especially long term. Long-term clinical trials are needed to confirm the safety, side effects, clinical significance, and relative efficacy of Epithalon therapy.
